Under the current meat sheep breeding system, newborn lambs usually live with their mothers until weaning, and in daily management, they often need to be separated from their ewes for a short period due to dehorning, disease treatment, etc. Such short-term separation was considered to be a high-intensity stress for the lambs. This study aimed to explore the effects of 1 h sensory separations on behaviors and the concentration of stress-related indicators of small-tailed Han lambs. Lambs were assigned to four groups: auditory, visual, and tactile separation (AVT) group; visual and tactile separation (VT) group; tactile separation (T) group; and control (C) group. Then they were separated from their mothers for one hour on postnatal days 14, 21, 28, 35 and 42. Results showed the separated lambs (AVT, VT, and T groups) spent less time lying down relaxing and more time looking around, exploring, vocalizing, and attempting to escape (P < 0.05). Lambs separated by lack of tactile contact only exhibited the most escaping and moving behavior. Twin-born lambs showed less moving, escaping, and vocalizing than single-born lambs (P < 0.05). The separation also led to a rise in serum globulin levels and a decrease in tetraiodothyronine. In conclusion, this study showed that temporary 1 h ewe-lamb separations could affect behaviors and the serum levels of stress indicators of lambs. The behavioral responses were more obvious when lambs were separated by lack of tactile contact only, and in single-born lambs. It can conclude that indicated that when lambs need to be temporarily separated from ewes in daily management production, it would be better to let them stay together with their littermates, and make them avoid hearing or seeing the ewes, such management may partially reduce the separation stress, thereby improving the welfare and breeding efficiency of sheep.

Inappropriate management practices of domestic animals during pregnancy can be potential stressors, resulting in complex behavioural, physiological and neurological consequences in the developing offspring. Some of these consequences can last into adulthood or propagate to subsequent generations. We systematically summarized the results of different experimental patterns using artificially increased maternal glucocorticoid levels or prenatal maternal physiological stress paradigms, mediators between prenatal maternal stress (PMS) and programming effects in the offspring and the effects of PMS on offspring phenotypes in sheep. PMS can impair birthweight, regulate the development of the hypothalamic-pituitary-adrenal axis, modify behavioural patterns and cognitive abilities and alter gene expression and brain morphology in offspring. Further research should focus on the effects of programming on gene expression, immune function, gut microbiome, sex-specific effects and maternal behaviour of offspring, especially comparative studies of gestational periods when PMS is applied, continual studies of programming effects on offspring and treatment strategies that effectively reverse the detrimental programming effects of prenatal stress.

In this study, the regulation effects of selenium (Se) on the expression of pyrin domain-containing protein (NLRP) 3 inflammasome and reactive oxygen species (ROS) in bovine mammary epithelial cells (bMECs) infected by Staphylococcus aureus (S. aureus) were detected. bMECs were treated with 8 µmol/L Na2SeO3 for 12 h before infection with S. aureus for 2 h. Through flow cytometry, Western blot, and qRT-PCR analysis, the expression of ROS and NLRP3 imflammasome was detected. Results showed Se significantly reduced the ROS level in bMECs; at the same time, the expressions of NLRP3, ASC, caspase-1, Pro-IL-1ß, and IL-1ß were also decreased. In conclusion, Se inhibits S. aureus-induced inflammation by suppressing the activation of NLRP3 inflammasome and ROS in bMECs.

Mounting evidence revealed the importance of gut microbiota in host metabolism, immunity and physiology, and health. Yimeng black goats (YBGs) mainly distributed in Shandong province of China, displayed a complicated intestinal microecosystem, but studies of its gut microbiota are still insufficient to report. Therefore, this study was performed with an objective to characterize the intestinal microbial community structure and diversity in the small intestine (duodenum, jejunum and ileum) and cecum of YBGs and investigated the variability of gut microbiota of different intestinal segments. A total of 12 intestinal samples were collected from YBGs for high-throughput sequencing analysis based on V3-V4 variable region of 16S rRNA genes. Our results revealed alterations in gut microbial composition with obvious differences in relative abundance between the different intestinal segments. Additionally, small intestine including duodenum, jejunum and ileum not only displayed higher species abundance and diversity than cecum but also showed a significant difference among the main components of gut microbiota based on the analytical results of alpha and beta diversities. At the phylum level, Firmicutes and Proteobacteria were the most preponderant phyla in all the samples regardless of intestinal sites. Moreover, the microbiota in small intestine was significantly different from cecum, which were characterized by the higher relative abundance of Butyrivibrio_2, Megasphaera, Halomonas, Delftia, Hydrogenophaga, Limnobacter, Pseudoxanthomonas, Novosphingobium, Janibacter and Erythrobacter, whereas the levels of Butyricicoccus, unidentified_Lachnospiraceae, Fusicatenibacter, Akkermansia, Ruminococcaceae_NK4A214_group and Lactobacillus were lower. Overall, this study first characterized the profile of gut microbiota composition in different intestinal sites and provide better insight into intestinal microbial community structure and diversity of YBGs.

BACKGROUND: Lucerne is a perennial legume forage, which can produce multiple cuts in 1 year. Microelements play fundamental roles in the function, maintenance and adaptation to the environment for lucerne growth. However, the role of the accumulation of copper (Cu), iron (Fe), manganese (Mn) and Zinc (Zn), which vary with lucerne ages or cuts, has not been previously determined. Therefore, a hypothesis on the Cu, Fe, Mn and Zn in lucerne varying with age and cut was tested. METHODS: A total of 11, 8, 5, 4 and 1 year old lucerne (Medicago sativa Longdong) were selected as the material (until 2012 year), and samples were taken as three cuts at the cutting periods (early flowering stage) in 2012. Then, the contents and yields of Cu, Fe, Mn and Zn in lucerne were measured and calculated. RESULTS: The highest contents of Cu, Fe, Mn and Zn in lucerne were found in the 1 year old among the five ages, at the 3rd cut compared to the other two cuts, and in the leaf among the three organs. The highest yields of Cu, Fe, Mn and Zn were found in the older ages (11 and 8 years old), at the 3rd cut, and in the root among the three organs. The most positive correlations were found between contents, yields and biomass. CONCLUSIONS: The hypothesis was supported by the results. And the contents and yields of lucerne Cu, Fe, Mn and Zn were affected by the age, cut and organ. Furthermore, the yields of lucerne Cu, Fe, Mn and Zn were determined by their contents and lucerne biomass.

BACKGROUND: Colonization of intestinal microbiota in ruminant during the early life is important to host health, metabolism and immunity. Accumulating evidence revealed the ameliorative effect of milk replacer administration in the gut microbial development of early-weaned ruminants. Yimeng black goats (YBGs) inhabiting Shandong, China show a complex intestinal microbial ecosystem, but studies of their gut microbiota are still insufficient to report. Here, this study was performed to investigate how the gut microbiota develops in weaned YBGs with the effect of age and milk replacer. RESULTS: Results indicated that both age and milk replacer were important factors to change the gut microbiota of YBGs. Although the alpha diversity of gut microbiota did not change with the age of YBGs, the taxonomic compositions significantly changed. The relative abundance of some beneficial bacteria such as Lachnospiraceae, Ruminococcaceae, Ruminiclostridium, Eubacterium and Barnesiella significantly decreased and subsequently increase with age, which contributes to maintain the stability of intestinal environment and realize the diversity of intestinal functions. The relative abundance of Porphyromonas, Brevundimonas, Flavobacterium, Stenotrophomonas, Propionibacterium, Acinetobacter, Enterococcus and Clostridium belong to pathogenic bacteria in milk replacer-treated YBGs was significantly decreased. Additionally, some beneficial bacteria such as Ruminococcus, Ruminococcaceae, Christensenellaceae and Ruminiclostridium also display a trend of decreasing first followed by gradually increasing. CONCLUSIONS: This study first revealed the gut bacterial community alterations in YBGs with the effect of age and milk replacer. This study also characterized the gut microbial distribution in YBGs with different ages and provided better insight into microbial population structure and diversity of YBGs. Moreover, milk replacer may serve as a good applicant for improving gut microbial development in early-weaned YBGs.

Increasing evidence has indicated the ameliorative effect of milk replacer supplementation in ruminants for regulating their early growth and rumen development. However, it is still unclear whether milk replacer supplementation has a beneficial role in the growth performance and rumen microbiota of Yimeng black goats (YBGs). Therefore, this study was performed to investigate the effects of milk replacer on growth performance and rumen microbiota of YBGs. Our results revealed that milk replacer supplementation could significantly improve the growth performance of YBGs. Additionally, the results of alpha and beta diversities indicated that there was no significant difference in richness and diversity between the control and milk replacer-treated YBGs. At the phylum level, Bacteroidetes, Firmicutes, and Proteobacteria were the most dominant phyla in all the samples at different stages. Moreover, the YBGs treated with milk replacer possessed a higher abundance of Verrucomicrobia than that in the control YBGs, while the level of Actinobacteria was obviously decreased. It is noteworthy that the abundance of Proteobacteria in the control YBGs was higher than that in the YBGs supplemented with milk replacer throughout the experiment. At the level of genus, the differences in the richness between control and milk replacer supplement YBGs were gradually observed. Compared with the control YBGs, the proportion of Akkermansia, Veillonella, Anaerovibrio, Ruminococcaceae_NK4A214_group, Ruminococcus_1, and Ruminococcus_2 was increased in the YBGs treated with milk replacer, whereas Turicibacter was decreased. In conclusion, milk replacer supplementation may serve as a good applicant for ameliorating early YBGs development and rumen microbiota.

Rutin, a widely distributed phytochemical flavonoid, can be used to control insect pests. In this study, we studied the growth performance of the grasshopper Oedaleus asiaticus Bey-Bienko given xenobiotic rutin using feeding experiments and transcriptomic analysis. O. asiaticus had reduced body size, lower survival rate, and reduced growth performance when fed with xenobiotic rutin. Rutin-fed nymphs had large variation in gene expression profiles, with a total of 308 genes significantly upregulated and 287 genes downregulated. The upregulated genes were significantly enriched in stress resistance-, immune-, and detoxification-related biological processes and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. Downregulated genes mainly involved cuticle biosynthesis and nutrition metabolism-related pathways. The quantitative real-time PCR (qRT-PCR) analysis of 15 candidate genes also produced results consistent with the transcriptome data. These results suggested that grasshoppers' capacity for biosynthesis and nutrition metabolism decreased, and stress resistance and metabolized capacity to toxic substances were significantly induced when O. asiaticus was fed on xenobiotic rutin. Rutin, as a phytotoxin, had detrimental effects and induced changes in gene expression profiles for O. asiaticus. This study can provide a molecular basis and offer future opportunities for the development of rutin-related insecticides and their application to grasshopper control.

The grasshopper Calliptamus abbreviatus Ikonn is a significant pest species distributed across the northern Asian grasslands. Grasshopper plagues often result in significant loss to plant biomass and subsequent deterioration of grass quality that leads to economic depletion. To better understand the close relationship between C. abbreviatus and host plant species, a 2-yr study was conducted. Results showed that the relative density of C. abbreviatus was positively correlated with aboveground biomass of the plant Artemisia frigida. We hypothesized that A. frigida, the most favorable food resource, was optimal for growth performance and that the presence of this plant species led to C. abbreviatus plagues. A controlled feeding trial showed that C. abbreviatus had better growth performance (i.e., survival rate, body mass, and growth rate) when fed on A. frigida and this host was preferred over other plant species since the consumption and food utilization efficiency on plant was comparatively greater. These results were consistent with the distribution of C. abbreviatus in the grassland and suggested that the presence of A. frigida significantly improved C. abbreviatus growth performance. These findings will be useful for designing improved pest management strategies in response to grassland vegetation succession due to grazing, climate change, or human interference.

Yimeng black goat is one of the national breeds of geographical indication in China and is one of the key protected local livestock and poultry breeds of Shandong province. The complete mitochondrial genome sequence of Yimeng black goat was investigated in this study (GenBank accession no. MT134111). The mitogenome (16,640 bp) consisted of a non-coding control region (D-loop region), two ribosomal RNA (rRNA) genes, 13 protein-coding genes (PCGs), and 22 transfer RNA (tRNA) genes. The complete mitochondrial genome sequence and the neighbour-joining tree of the Yimeng black goat would contribute to further study in genetic mechanism and phylogenomic research of goats.

Diets essentially affect the ecological distribution of insects, and may contribute to or even accelerate pest plague outbreaks. The grasshopper, Oedaleus asiaticus B-Bienko (OA), is a persistent pest occurring in northern Asian grasslands. Migration and plague of this grasshopper is tightly related to two specific food plants, Stipa krylovii Roshev and Leymus chinensis (Trin.) Tzvel. However, how these diets regulate and contribute to plague is not clearly understood. Ecological studies have shown that L. chinensis is detrimental to OA growth due to the presence of high secondary metabolites, and that S. krylovii is beneficial because of the low levels of secondary metabolites. Moreover, in field habitats consisting mainly of these two grasses, OA density has negative correlation to high secondary metabolites and a positive correlation to nutrition content for high energy demand. These two grasses act as a 'push-pull,' thus enabling the grasshopper plague. Molecular analysis showed that gene expression and protein phosphorylation level of the IGF → FOXO cascade in the insulin-like signaling pathway (ILP) of OA negatively correlated to dietary secondary metabolites. High secondary metabolites in L. chinensis down-regulates the ILP pathway that generally is detrimental to insect survival and growth, and benefits insect detoxification with high energy cost. The changed ILP could explain the poor growth of grasshoppers and fewer distributions in the presence of L. chinensis. Plants can substantially affect grasshopper gene expression, protein function, growth, and ecological distribution. Down-regulation of grasshopper ILP due to diet stress caused by high secondary metabolites containing plants, such as L. chinensis, results in poor grasshopper growth and consequently drives grasshopper migration to preferable diet, such as S. krylovii, thus contributing to grasshopper plague outbreaks.

BACKGROUND: Sheep have developed the ability to store fat in their tails, which is a unique way of reserving energy to survive a harsh environment. However, the mechanism underlying this adaptive trait remains largely unsolved. RESULTS: In the present study, we provide evidence for the genetic determinants of fat tails, based on whole genome sequences of 89 individual sheep. A genome-wide scan of selective sweep identified several candidate loci including a region at chromosome 13, a haplotype of which underwent rapid evolution and spread through fat-tailed populations in China and the Middle East. Sequence analysis revealed an inter-genic origin of this locus, which later became a hotspot of ruminant-specific retro-transposon named BovB. Additionally, the candidate locus was validated based on a fat- and thin-tailed cross population. The expression of an upstream gene BMP2 was differentially regulated between fat-tailed and thin-tailed individuals in tail adipose and several other tissue types. CONCLUSIONS: Our findings suggest the fixation of fat tails in domestic sheep is caused by a selective sweep near a retro-transposable hotspot at chromosome 13, the diversity of which specifically affects the expression of BMP2. The present study has shed light onto the understanding of fat metabolism.

Background: Animal domestication has been extensively studied, but the process of feralization remains poorly understood. Results: Here, we performed whole-genome sequencing of 99 sheep and identified a primary genetic divergence between 2 heterogeneous populations in the Tibetan Plateau, including 1 semi-feral lineage. Selective sweep and candidate gene analysis revealed local adaptations of these sheep associated with sensory perception, muscle strength, eating habit, mating process, and aggressive behavior. In particular, a horn-related gene, RXFP2, showed signs of rapid evolution specifically in the semi-feral breeds. A unique haplotype and repressed horn-related tissue expression of RXFP2 were correlated with higher horn length, as well as spiral and horizontally extended horn shape. Conclusions: Semi-feralization has an extensive impact on diverse phenotypic traits of sheep. By acquiring features like those of their wild ancestors, semi-feral sheep were able to regain fitness while in frequent contact with wild surroundings and rare human interventions. This study provides a new insight into the evolution of domestic animals when human interventions are no longer dominant.

The effects of parity and litter size on maternal behavior of Small Tail Han sheep was investigated at Linyi University, China. Sixty-eight ewes were observed from parturition to weaning. Continuous focal animal sampling was used to quantify the duration of maternal behaviors. Ewe feces were collected every 2 days and estradiol concentration was measured with an enzyme immunoassay kit. All lambs were weighed 24 h after parturition and again at 35 days of age. Parity increased sucking, following, grooming, low-pitched bleat, head-up and udder-refusal behavior and decreased aggressive behavior (P < 0.01, P < 0.01, P < 0.05, P < 0.05, P < 0.05, P < 0.05, P < 0.01, respectively), and litter size showed significant effect on sucking, following and low-pitched bleat behavior (P < 0.05, P < 0.01, P < 0.05, respectively). The lambs of multiparous ewes were significantly heavier than primiparous ewes at birth (P < 0.01) and were significantly heavier at weaning age (P < 0.01). Similar results were founded for birth weight and weaning weight gain in litter size (P < 0.01, P < 0.01, respectively). Estradiol concentration in feces was higher in multiparous ewes than primiparous ewes. Parity and litter size may have effects on maternal behavior during lactation. Ewes that have 2-3 lambs may be more suitable for production of Small Tail Han sheep in China.
